Yeah the cartridge is permanent, non removable.

As poll said it looks like your jumper wire is in the incorrect spot. This is taken directly from the wiki. With my current and first GBZ I'm using the powerboost 1000c and I'd like some status leds on the outside of the case so I plan to solder external leds from the powerboost ones. However ...

If it's a fresh install of retropie your using the default of the audio settings is set to something like 50%. Go to retropie configuration then audio, then mixer - adjust output volume. Turn it to max then you can use your volume wheel to adjust.
